Prototype and Scriptaculous are the bomb. If you're not using them yet in all your sites, you should be.

The trick is to plan ahead for the endgame. Each move earns you points equivalent to the (mathematical) square of the number of (colored) squares that disappear with that move. So it's best to sacrifice small groups at the beginning if it means you'll be able to remove groups of twenty-five adjacent squares at the end.
